Background

Motor neuron (motor neuron) diseases such as amyotrophic lateral sclerosis (amyotrophic lateral sclerosis) are characterized by the loss of cortical and spinal MNs.

Purpose

To study the effects of decreased GABA synthesis on spinal cord motor function, we analyzed the effects of administering three selective inhibitors of glutamate decarboxylase: 3-mercaptopropionic acid (mercaptopropionic acid), a competitive inhibitor of glutamate decarboxylase, thiosemicarbazide (thiosemicarbazide), and pyridoxal phosphate γ-glutamyl hydrazone (PLPGH), two glutamate decarboxylase cofactor blockers.

GABA synthesis depends on the activity of the enzyme glutamate decarboxylase (glutamate decarboxylase), of which two isoforms are known: GAD65 and GAD67.

A single dose of any of these inhibitors did not affect motor behavior or motor neuron morphology.

Conclusion

These findings suggest that chronic inhibition of glutamate decarboxylase can lead to motor neuron degeneration and further suggest that GABA metabolism in the spinal cord may participate in the mechanisms that cause motor neuron death in patients with neurodegenerative diseases such as amyotrophic lateral sclerosis.
