Background

Prostate-specific membrane antigen (prostate-specific membrane antigen)-targeted molecular imaging has greatly improved the precision of prostate cancer (prostate cancer) diagnosis; however, the integration of high-sensitivity positron emission tomography (positron emission tomography) and high-resolution magnetic resonance imaging (magnetic resonance imaging) into a single, safe nanoplatform remains a critical clinical need.

Here, we report prostate-specific membrane antigen-targeted positron emission tomography/MR dual-modal ultrasmall manganese ferrite nanoparticles (ultrasmall manganese ferrite nanoparticles) for molecular imaging of prostate cancer.

Uniform ultrasmall manganese ferrite nanoparticles were synthesized via a dynamic simultaneous thermal decomposition (dynamic simultaneous thermal decomposition) method and their surfaces were sequentially engineered with the highly specific prostate-specific membrane antigen ligand glutamic acid-urea-lysine (Glu-urea-Lys) and the chelator NOTA, followed by efficient radiolabeling with 68 Ga.

Conclusion

This work demonstrates that combining the inherent MR T 1 contrast enhancement capability and favorable pharmacological properties of ultrasmall manganese ferrite nanoparticles with prostate-specific membrane antigen targeting and stable 68 Ga labeling yields a highly specific, safe, and quantitative positron emission tomography/MR nanoplatform, offering substantial promise as a potential clinically translatable platform for precise molecular imaging and personalized management of prostate cancer.
